Introduction of Affinity Chromatography

Affinity chromatography (AC), the most selective and versatile of the liquid chromatography methods, relies on proteins and their ligands such as hormones and their receptors, enzymes and their substrates, antibodies, and antigens, etc.) for reversible site-specific binding. Interactions can be used to immobilize a phase of interacting species on a column as a stationary phase for AC. The protein of interest is eluted from the complex fraction by selective adsorption of the affinity ligand. AC has become the most specific and effective chromatographic technique for separating and purifying biological macromolecules such as proteins and enzymes, and it has been widely used in biological separation.
