Size Exclusion Chromatography

Introduction of Size Exclusion Chromatography

Molecular weight polydispersity is one of the basic characteristics of polymers. The properties of a polymer are closely related to its molecular weight and molecular weight distribution. Size exclusion chromatography (SEC) is a branch of liquid chromatography and has become an important means to determine the molecular weight distribution and structure of polymers and reveal the physical properties of polymer samples. In addition, it plays an active role in the quality control of polymer products and the improvement of synthesis processes. SEC methods are generally divided into two categories according to their elution systems, namely gel permeation chromatography (GPC) and gel filtration chromatography (GFC).

Larger particles take a more direct path through the gel and elute first.Figure. 1 Larger particles take a more direct path through the gel and elute first. (Liu D S K, et al., 2020)

Principle of SEC

The principle of SEC separation is to separate substances according to their size (molecular weight). High molecular weight components cannot enter the pores of the stationary phase and flow out first; medium molecular weight components can enter part of the pores of the stationary phase and then flow out; low molecular weight components can enter all pores and flow out last.

Types of Our Products

Outstanding Advantages of Our SEC Products

  • Peak appear quickly
  • High activity recovery rate
  • No impurity or residue will be retained on the chromatographic column
  • Accurate determination of molecular weight and molecular weight distribution
  • Large column loading capacity
  • The solute is independent between the stationary phase and the mobile phase without gradient elution

SEC has been used for quality control of biological products since the early development of recombinant proteins for pharmaceutical use. Furthermore, the technology has been continuously improved and is now well-used for protein characterization.

Application of Our SEC Products

  • Standard separations for quantifying protein dimers, trimers, and oligomers
  • Calibration of complex polymers
  • Biopharmaceutical characterization
  • Routine monitoring of aggregation and quaternary structure of modified proteins (protein-drug conjugates and pegylated proteins)
  • Guide the development of biopharmaceutical purification processes
  • Changes in the extent and form of protein aggregation
