Alpha-Chymotrypsin is a serine endoprotease that preferentially targets the C-terminal peptide bonds of large hydrophobic residues, including Tyr, Phe, Trp, and Leu. Secondary cleavage may occur at Met, Ala, Asp, and Glu. During production, TLCK is utilized to neutralize residual trypsin activity, while calcium ions are added to enhance the enzyme's stability and activation.

Usage Notes
Apply α-chymotrypsin at a 1:50 (w/w) protease-to-protein loading. The digestion should proceed in a 100 mM Tris-HCl buffer (pH 7.8) supplemented with 10 mM CaCl2. Incubate at 30 °C for a duration of 2 to 12 hours.
