Cat#No# | GC-364C |
Product Overview | GCSFSelect is an affinity resin designed for the purification of G-CSF products, rendering high purity and yield in one step: Rigid agarose base matrix allows high flow rates and processing of large sample volumes for increased throughput. Non-mammalian derived product reduces regulatory concerns in the production of G-CSF for clinical applications. |
Description | G-CSF (granulocyte-colony stimulating factor) is a hormone stimulating the bone marrow to produce more white blood cells. The ligand was developed based on the BAC B.V. technology and specifically screened to bind and elute G-CSF targets in the presence of other proteins from different expression systems. GCSFSelect is an affinity resin designed for the purification of G-CSF (granulocyte-colony stimulating factor) products, enabling high yield and purity in one step. |
Characteristic | Efficient, industrial-scale purification of GCSF by affinity chromatography. High flow rates for processing of large sample volumes to increase throughput. Reduced regulatory concerns (due to non-mammalian derived product) in the production of GCSF for clinical applications. |
Maximum operating pressure | 300 kPa at 600 cm/h, 1 m diameter column, 20 cm bed height |
Matrix | Highly cross-linked high-flow agarose |
Average particle size | ~75 µm |
Ligand | Recombinant protein (Mr 14 400), produced in S. cerevisiae, with affinity for the protein G-CSF. |
Ligand density | Approx. 7,5 mg/ml of medium |
Dynamic binding capacity | Approx. 3.9 mg GCSF/mL resin |
Recommended flow rate | Minimum 600 cm/h in a 1 m diameter column, with 20 cm bed height at 20°C using buffers with the same viscosity as water at < 0.3 MPa (3 bar) |
Recommended column height | 20 cm |
pH working range | 3–10 |
CIP stability | 2–12 |
Temperature stability | 4°C to 30°C |
Storage | 20% ethanol at 2°C to 8°C. |
Cleaning-in-place | The recommended protocol comprises initial strip of the resin at low pH, and then subjecting the resin to NaOH of low concentration for cleaning. Lastly, PAB (120 mM phosphoric acid, 167 mM acetic acid, 2.2 % v/v benzyl alcohol) is used for final sanitization of the resin. |
Sanitization | PAB solution is sensitive to light and should be freshly made not to damage the resin. PAB solution should be stored in a dark bottle and kept no longer than for a week. PAB solution has a pH of < 2, and resin stability can be limited in prolonged exposure at such a low pH. |
