| Cat#No# | Hi-134P |
| Product Overview | HiPrep 16/10 Phenyl HP is a prepacked column designed for preparative hydrophobic interaction chromatography (HIC) with good resolution. |
| Description | Phenyl Sepharose HP chromatography resin is based on a matrix of highly cross-linked agarose beads, which have been derivatized with phenyl groups. These are attached to the agarose matrix via uncharged, chemically stable ether linkages. The result is a truly hydrophobic resin with no ionic properties. |
| Characteristic | Aromatic HIC resin for intermediate purification and polishing. Selectivity similar to Phenyl Sepharose 6 Fast Flow (Low Sub) but higher resolution due to the smaller 34 µm bead size. Provides fast, easy high-capacity separations in a convenient format. |
| Maximum operating pressure | 3 bar [0.3 MPa] (44 psi) |
| Matrix | Cross-linked agarose, spherical |
| Average particle size | ~ 34 µm |
| Ligand | Phenyl |
| Ligand density | ~ 25 µmol/mL resin |
| Recommended flow rate | < 150 cm/h |
| Recommended column height | 100 mm |
| Chemical stability | Stable to commonly used aqueous buffers, 0.01 M NaOH, 8 M urea, 6 M guanidine hydrochloride, 30% isopropanol, and 70% ethanol. |
| pH working range | 3 to 12 |
| CIP stability | 3 to 12 |
| Storage | 4 to 30°C, 0.2 M Sodium Acetate in 20% Ethanol |
| Cleaning-in-place | 80 mL of a 1.0 M NaOH (0.01 M NaOH for Phenyl Sepharose HP) solution (removes precipitated proteins, hydrophobically bound proteins, and lipoproteins from the column) followed by 80 mL distilled water. 80 mL of 70% ethanol or 30% isopropanol (removes proteins, lipoproteins, and lipids that are strongly hydrophobically bound to the column) followed by 60 mL distilled water. |
