Lys-N is a recombinant metalloendopeptidase that specifically hydrolyzes peptide bonds at the N-terminus of lysine residues. It is an essential tool for PTM research and de novo sequencing, as it facilitates the generation of b-ion-rich spectra. Pairing Lys-N with rTrypsin-N offers a comprehensive strategy for full sequence coverage.
Physical Appearance
Lyophilized powder
Molecular Weight
18.4 kDa
Resuspension Buffer
Utilize either Zinc Sulfate or Zinc Acetate buffer (50 μM).
Storage
Storage at –20 °C for powder; for the reconstituted enzyme, store at -80 °C for up to 30 days.
Shelf life
Store at -80 ℃ for up to 24 months.
Stability
Reaches full enzymatic potential within the pH 7.0 to 9.0 interval.
Usage Notes
Dissolve 20 μg of MS-grade Lys-N in 40 μL of buffer to activate. It is recommended to use Tris-HCl (pH 8.0) or 50 mM ammonium bicarbonate for the sample. Use a 1:30 enzyme-to-substrate ratio (with 0.5 μg/μL Lys-N) and incubate at 37 °C for 4 hours.
