rTrypsin-N is a novel recombinant protease that cleaves peptide bonds at the amino (N-terminal) side of lysine and arginine. This "reverse" specificity is particularly advantageous for protein sequencing and PTM localization. Peptides produced by rTrypsin-N typically yield a higher frequency of b-ions during mass spec fragmentation.

rTrypsin-N demonstrates peak catalytic activity at a neutral pH of 7.5.
Usage Notes
To ensure full potency, reconstitute MS-grade rTrypsin-N in 200 μL distilled water (pH 7.5). Cleavage is best performed in Tris-HCl (pH 7.5), 0.1% BT Surfactant, or 20–50 mM HEPES. Introduce the enzyme (0.5 μg/μL) at a 1:30 ratio to the substrate. Incubate the mixture for 4 hours at 37 °C.
