Description This optimized blend combines rTrypsin 3.0 and rLys C to ensure comprehensive protein digestion. By targeting both Lys/Arg (via Trypsin) and specifically Lys (via Lys-C), the mix significantly reduces missed cleavages caused by proline interference, PTMs, or C-terminal hydrophobicity, making it a gold standard for precise proteomic analysis.

Stability Maximum efficiency for the rTrypsin 3.0/rLys C blend is observed in the pH 7.0–9.0 range.
Usage Notes 1. Buffer options: 20 mM Tris, HEPES, or 50 mM ABC within pH 7.0–9.0. 2. For denatured substrates, apply a 1:50 protease-to-protein ratio. Ensure a 0.5 µg/µL protein concentration and incubate for 30 minutes at 37 °C.
Activity • rTrypsin: 15,000 units/mg. • rLys C: 25 units/mg.
Unit Definition Activity is defined by the hydrolysis of 1.0 µmol of BAEE/min (pH 7.8, 25°C, 253 nm) or 1.0 µmol of T-G-P Lys Nitroanilide/min (pH 7.8, 25°C, 405 nm).
