Chirality is a common phenomenon in nature, and the basic substances that constitute living organisms, such as most amino acids and sugars, are chiral molecules. Drug enantiomers in the body in most cases will show different therapeutic effects and even adverse reactions. Research on chiral drugs is the development trend of pharmaceutical chemistry. Using a single isomer to replace the mixed drug used in clinical applications is also one of the ways to develop new drugs. With the deepening understanding of chiral molecules, the demand for single chiral substances is increasing, and the requirements for its purity are getting higher and higher. The preparation of chiral drugs or intermediates by chiral separation of racemes obtained by ordinary chemical synthesis is the main method for the industrial production of chiral drugs.

Supercritical fluid chromatography (SFC) has been widely used in small molecule drug development strategies by various drug research and development institutions. It is certain that SFC will be applied in the development process of chiral candidate drugs for a long time, including the screening of chiral analysis methods for determining API and chiral purity of intermediates. As well as the development of chiral separation methods scaling up. The prep-SFC can be used for chiral separation from milligram to kilogram. In many cases, the yield of SFC purified samples from the same library is higher than that of HPLC. (Fig. 1) The low viscosity and high diffusivity of the mobile phase in the SFC allow for higher flow rates and lower pressure drops compared to traditional HPLC. In addition, SFC requires less organic or aqueous organic solvents than LC and provides another normal phase retention mechanism. (Pinkston J. D., et al, 2006)
