Ligand Immobilization

NHS Ester-activated Supports

NHS ester is a reactive group formed by EDC activation of carboxylate molecule. NHS ester-activated resins react with primary amines in alkalescency conditions (pH 7.2-8.5) to form stable amide bonds, which leads to immobilization of the ligands.

Aldehyde-activated Supports

Reductive amination is a reliable and popular chemical process for covalently attaching antibodies or other proteins to beaded agarose resins. The reaction begins with the formation of an initial Schiff base between an aldehyde and an amine group, followed by the addition of NaCNBH3 to reduce to a secondary amine. Depending on the type and amounts of ligands involved, the coupling efficiency of the reaction can be more than 85%.
